In Peru, the land of the Incas, the Indian pin their hopes on Jaucarapora. They want to him as their king who could drive back the white conquistador. The president gives him four weeks to prove his innocence. No matter where the Jaguar goes, the craftily circulated news that he is wanted killer is always there ahead of him. Graciela, his dead brother's daughter, sets out alone to forewarn him. The Jaguar finds out that Gambusino has killed his brother and Jinaucarapora's father. Jaucarapora meets Graciela and saves her life. They fall in love each other. Gambusino tries to kill the Jaguar but perishes himself. Graciela persuades Jaucarapora that an uprising against the incomparably stronger whites will ruin his people for good. He rejects the sacred crown. The last Inca suffers death at the hands of his own mentor.

In Arizona Territory, saloon owner Jennie Lee is facing a hostile takeover from her business competitor and town strong man Zack Williams. At the same time, silver mine owner Clementine Hewitt has problems with Mexican bandits who regularly rob her shipments of silver leaving the mine for the nearest town bank. Clementine suspects that Zack Williams is the mastermind hiring the Mexican bandits but she cannot prove it. Fed up with the whole situation, Jennie Lee contacts her old friend, Wyatt Earp, and asks him to come to Rio Bravo and render assistance. He arrives in town under the guise of a drifter named Laramie. Once in town, Laramie meets with Jennie Lee at the saloon and after being briefed he starts snooping around. The otherwise indifferent town sheriff, Leo Wynn, who hates Williams' grip on the territory, decides to help Laramie. During his investigation, Laramie is frequently attacked, inside and outside the town, by Williams' henchmen and by the Mexican bandits led by desperado Pancho Bogan. Williams even tries to frame Laramie up for the murder of one of Clementine Hewitt's mine employees. Sheriff Leo Wynn doesn't buy it. With every passing day Laramie gets closer to the truth.
